在正常细胞中，磷脂酰丝氨酸（PS）只分布在细胞膜脂质双层的内侧，而在细胞凋亡早期，细胞膜中的磷脂酰丝氨酸（PS）由脂膜内侧翻向外侧。Annexin V 是一种分子量为 35~36kD 的 Ca2+依赖性磷脂结合蛋白，与磷脂酰丝氨酸有高度亲和力，故可通过细胞外侧暴露的磷脂酰丝氨酸与凋亡早期细胞的胞膜结合。因此 Annexin V 被作为检测细胞早期凋亡的灵敏指标之一。将 Annexin V 进行荧光素 PE 标记，以标记了的Annexin V 作为荧光探针，利用荧光显微镜或流式细胞仪可检测细胞凋亡的发生。7-AAD（7-amino-actinomycin D）是一种核酸染料，它不能通过正常质膜，随着细胞凋亡、细胞死亡过程，质膜对 7-AAD 的通透性逐渐增加，结合细胞凋亡中 DNA 的有控降解，在合适波长激发光的激发下可发出明亮的红色荧光，通过 7-AAD 标记 DNA 的强弱，将细胞分为三群：7-AAD 强为死亡细胞，7-AAD 弱是凋亡细胞，7-AAD-为正常活力细胞。7-AAD 同 PI 有着相似的荧光特性，但其发射波谱较 PI 窄，对其他检测通道的干扰更小，在多色荧光分析中是 PI 的理想替代品，可与 Annexin V-PE 联合使用。本试剂盒可应用于培养细胞凋亡检测（不推荐用于检测组织样本）。
